Supportive Studies: Tissue Banks
Delayed immune reconstitution (IR) following HCT results in prolonged susceptibility to infection and is a major cause of morbidity and mortality. The rate of IR is directly correlated with the number of hematopoietic stem cells (HSC) infused and is particularly delayed in patients undergoing cord blood transplantation (CBT) secondary to the limited numbers of HSC.
We are interested in studying the IR of CB progenitors in mice. However, human thymopoiesis in a mouse thymus is inefficient and the MHC restriction and tolerance of human T cells that are selected in mouse thymus is unknown. Thus, we will evaluate CB engraftment in a human thymus xenograft mouse model (huNOD/SCID), in which immunodeficient mice are implanted with human thymic tissue beneath the kidney capsule. Once established, the thymic organ successfully reproduces the differentiation, proliferation and function of human HSC progenitors within the context of the human thymus.Thus, the hu NOD/SCID model is superior for evaluation of human T cell reconstitution compared to other mouse models.
