The second area of interest of the laboratory involves the study of the zinc-finger transcription factor GATA-2. GATA-2 is expressed at very high levels in primitive hematopoietic stem cells and expression dramatically decreases with hematopoietic cell maturation. We have observed that enforcement of the normally high level of GATA-2 n stem cells, using retroviral vectors expressing GATA-2, blocks the ability of stem cells to participate in hematopoiesis in vivo. Future experiments are aimed at elucidating the mechanism of this functional stem cell "arrest" through the use of microarray technology and conditionally expressing transgenic mice.
