Labeled Mass Spectral Library (Version: v1)

Introduction

This page provides both human and mouse tandem mass tag (TMT) labeled spectral libraries for download. The spectra were acquired by either Q Exactive HF or Orbitrap Fusion at St. Jude Proteomics Core over years. Human and mouse libraries were derived from 16 and 26 LC-MS/MS runs, and 863 and 1,498 fractions, respectively.

Briefly, the libraries were constructed with the following four steps. (1) A protein mixture sample is prepared (digested with trypsin, desalted, labeled, and separated by chromatography); (2) The fractionated samples are analyzed by a liquid chromatography tandem mass spectrometer (LC-MS/MS; either HF or Fusion); (3) The resulting MS/MS spectra are compared to theoretical product ions and then filtered at protein FDR < 1% using JUMP program (Wang et al., 2014); (4) The identified peptides and raw MS data were used to generate the library with SpectraST (Lam et al., 2006).
