Numerous studies have led to a model that human AML is initiated by pre-leukemic stem cells (pHSCs) and subsequently driven by leukemia stem cells (LSCs). However, few functional studies have been conducted in patient cells limiting the utility of these models for translational studies. This presentation will describe newly developed methods to interrogate important questions in both pHSCs and LSCs including the requirement for specific mutations in disease initiation and maintenance, the dynamics of individual LSCs, and the use of single cell multiomics to interrogate these populations.Â
